Summary:
Introduction. The problem of the spread of borderline mental disorders in pediatric practice continues to be relevant. Enlarged intellectual loads lead to emotional lability of children, their increased anxiety, sensitivity, psycho-emotional exhaustion, while working capacity decreases. In this case, drug prevention with the appointment of nootropics is relevant. Glycine was chosen as the object of study, which has advantages when used in pediatric practice, has a universal anti-stress effect, normalizes the state of the nervous system, has a mild sedative effect, improves mental performance and memory. Important properties of glycine are the absence of contraindications and side effects of the drug. The optimal dosage form for use in pediatrics for children are suppositories. The aim. The aim of our research work was the development of the composition of glycine suppositories for use in pediatric practice. Materials and methods. To achieve this goal, a comprehensive study was carried out, including physicochemical and biopharmaceutical studies. Initially, the selection of the optimal bases for the manufacture of suppositories was carried out. The melting point of hydrophobic bases and the dissolution time of hydrophilic bases have been studied. The degree of pharmaceutical availability of glycine from suppositories was assessed by the method of equilibrium dialysis through a semipermeable membrane, which was used as unvarnished cellophane, followed by spectrophotometric determination of the amount of released glycine. Results. Studies have been carried out on the development of glycine suppositories for use in pediatric practice. The selection of the therapeutic dose of glycine in the dosage form and the basis for suppositories was carried out. Glycine in the composition of hydrophobic suppository bases was introduced as a suspension. The pharmaceutical availability of glycine from the developed dosage form was studied by dialysis through a semipermeable membrane. Discussions. The maximum release of glycine from suppositories is provided by a base consisting of an alloy of cocoa butter with T-2 emulsifier. Studied the stability of the resulting suppositories during storage. Conclusion. The composition of suppositories with glycine was developed. The maximum pharmaceutical availability of glycine from suppositories is provided by a base consisting of 95% cocoa butter and 5% T-2 emulsifier.
